Amine acetylation is a diverse topic with importance to the regulation of several physiological processes as well as the metabolism of drugs and environmental chemicals. Arylalkylamine N‐acetyltransferase is widely distributed in several species, where this enzyme plays an important role in the seasonal regulation of reproduction and photoperiodism in vertebrates through the pathway of melatonin formation. In insects, this enzyme is involved in monoamine neurotransmitter inactivation and the formation of catecholamine intermediates necessary for sclerotization of the insect cuticle.

This study used fMRI to investigate the neural substrates of moral cognition in health resource allocation decision problems. In particular, it investigated the cognitive and emotional processes that underpin utilitarian approaches to health care rationing such as Quality Adjusted Life Years. Participants viewed hypothetical medical and nonmedical resource allocation scenarios which described equal or unequal allocation of resources to different groups. In addition, participants were assigned to 1 of 2 treatments in which they either did or did not receive advanced (...) instructions about the principles of utilitarianism. In all cases, participants were asked to judged the proposed allocations as “fair” or “unfair.” More brain activity was observed within the superior parietal lobe, angular gyrus, middle temporal gyrus, and bilateral caudate nucleus when participants viewed scenarios depicting equal divisions of resources. Conversely, unequal resource divisions were associated with more activity in the inferior frontal gyrus and insula cortex. Furthermore, instructions about the principles of utilitarianism led to significant activation differences within the inferior frontal gyrus and the middle frontal gyrus. Significant differences in activity were also found within the inferior frontal cortex and anterior insula between medical and nonmedical scenarios. The implications for cognitive control mechanisms and the cognitive and neural bases of utilitarian ethical judgment are discussed. (shrink)

This collection of essays, offered in honor of the distinguished career of prominent political philosophy professor Clifford Orwin, brings together internationally renowned scholars to provide a wide context and discuss various aspects of the virtue of “humanity” through the history of political philosophy.

We have synthesized a 582,970-base pair Mycoplasma genitalium genome. This synthetic genome, named M. genitalium JCVI-1.0, contains all the genes of wild-type M. genitalium G37 except MG408, which was disrupted by an antibiotic marker to block pathogenicity and to allow for selection. To identify the genome as synthetic, we inserted "watermarks" at intergenic sites known to tolerate transposon insertions. Overlapping "cassettes" of 5 to 7 kilobases (kb), assembled from chemically synthesized oligonucleotides, were joined by in vitro recombination to produce intermediate (...) assemblies of approximately 24 kb, 72 kb ("1/8 genome"), and 144 kb ("1/4 genome"), which were all cloned as bacterial artificial chromosomes in Escherichia coli. Most of these intermediate clones were sequenced, and clones of all four 1/4 genomes with the correct sequence were identified. The complete synthetic genome was assembled by transformation-associated recombination cloning in the yeast Saccharomyces cerevisiae, then isolated and sequenced. A clone with the correct sequence was identified. The methods described here will be generally useful for constructing large DNA molecules from chemically synthesized pieces and also from combinations of natural and synthetic DNA segments. 10.1126/science.1151721. Pancreatitis is a complex, progressively destructive inflammatory disorder. Alcohol was long thought to be the primary causative agent, but genetic contributions have been of interest since the discovery that rare PRSS1, CFTR and SPINK1 variants were associated with pancreatitis risk. We now report two associations at genome-wide significance identified and replicated at PRSS1-PRSS2 and X-linked CLDN2 through a two-stage genome-wide study. The PRSS1 variant likely affects disease susceptibility by altering expression of the primary trypsinogen gene. The CLDN2 risk allele is (...) associated with atypical localization of claudin-2 in pancreatic acinar cells. The homozygous CLDN2 genotype confers the greatest risk, and its alleles interact with alcohol consumption to amplify risk. It remains unclear whether such genetic variants are associated with disease aggressiveness. The NCI-SPORE Genetics Working Group retrospectively collected clinicopathologic information and genotype data for 36 SNPs which at the time had been validated to be associated with PC risk from 25,674 cases with PC. Cases were grouped according to race, Gleason score and aggressiveness. Statistical analyses were used to compare the frequency (...) of the SNPs between different disease cohorts. After adjusting for multiple testing, only PC-risk SNP rs2735839 was significantly and inversely associated with aggressive and high-grade disease in European men. Similar associations with aggressive and high-grade disease were documented in African-American subjects. The G allele of rs2735839 was associated with disease aggressiveness even at low PSA levels in both European and African-American men. Our results provide further support that a PC-risk SNP rs2735839 near the KLK3 gene on chromosome 19q13 may be associated with aggressive and high-grade PC. Future prospectively designed, case-case GWAS are needed to identify additional SNPs associated with PC aggressiveness. (shrink)

Experience has recently reemerged as an important analytical category for historians of the Old Regime and the French Revolution. Reacting against the perceived excesses of discourse analysis, which made political language independent of any social determinants, certain post-revisionists are now seeking to contextualize political language by relating it to the experience of those who use it. Political agency, in these analyses, is understood to be the effect of particular formative experiences. This article suggests that the search for an experiential antidote (...) to discourse is misconceived because it perpetuates an untenable dichotomy between thought and reality. Access to the phenomenon of historical agency should be pursued not through experience or discourse but through the category of consciousness, since the make-up of the subject's consciousness determines how he/she engages the world and decides to attempt changing it. After a brief discussion of an important study that exemplifies both the allure and the functionality of the notion of experience, Timothy Tackett's Becoming a Revolutionary, the article focuses on the evolving political consciousness of a man who became a revolutionary agitator in 1789, J.-M.-A. Servan. Analysis of his writings between 1769 and 1789 shows that the way in which his perspective was constructed, rather than the lessons of experience per se, determined the shape of his revolutionary intentions in 1789. Is a coherent worldview that embraces both classical Christology and modern evolutionary biology possible? This article addresses a difficult case at the intersection of philosophical theology and truthmaker theory. I show that three views, together, lead to difficultiesin providing truthmakers for truths of contingent predication, such as that the bread is white. These three views are: the Catholic dogma of transubstantiation, astandard truthmaker theory, and a trope (or accident) view of properties. I present and explain each of these three views, at each step noting their connections to the thought of St. Thomas Aquinas. After (...) presenting the three views, I show why they entail a difficulty for providing truthmakers for truths of contingent predication,drawing on two cases that are not impossible, for all we know. I then present four ways that one can respond to this difficulty, afterward noting some shortcomingsof those responses. (shrink)

The doctrine of the Incarnation, that Jesus Christ was both truly God and truly human, is the foundation and cornerstone of traditional Christian theism. And yet this traditional teaching appears to verge on incoherence. How can one person be both God, having all the perfections of divinity, and human, having all the limitations of humanity? This is the fundamental philosophical problem of the Incarnation. Perhaps a solution is found in an analysis of what the traditional teaching meant by person, divinity, (...) and humanity, or in understanding how divinity and humanity were united in a single person. This Element presents that traditional teaching, then returns to the incoherence problem to showcase various solutions offered to it. (shrink)

Moral education requires interdisciplinary engagement across philosophy, psychology, and education. Positive psychologists regularly acknowledge the breadth and depth of wisdom regarding the cultivation of virtues present in philosophical and religious texts and consult such writings when creating constructs, but they are less prone to integrate scientific findings with historical texts as inquiry proceeds. Thus, we provide a comparative analysis of the advice given in Lorenzo Scupoli’s The Spiritual Combat, from traditional Christian moral wisdom literature and the research findings from positive (...) psychology on the cultivation of the virtue of patience. Points of convergence relate to the utility of engaging activities that include cognitive reappraisal, habit formation through daily practice, activating positive motivation, prayer, mantra/transcendental meditation, and cultivating elevation through meditation on moral exemplars. Areas of divergence include the advisability of suppression, the role of motivation, and necessity of spiritual intervention, which suggest areas for future inquiry in moral education. (shrink)

At any given time, a subject has a multiplicity of conscious experiences. A subject might simultaneously have visual experiences of a red book and a green tree, auditory experiences of birds singing, bodily sensations of a faint hunger and a sharp pain in the shoulder, the emotional experience of a certain melancholy, while having a stream of conscious thoughts about the nature of reality. These experiences are distinct from each other: a subject could experience the red book without the singing (...) birds, and could experience the singing birds without the red book. But at the same time, the experiences seem to be tied together in a deep way. They seem to be unified, by being aspects of a single encompassing state of consciousness. (shrink)

In this paper, I provide an overview of the Christian moral wisdom with respect to virtue formation and character cultivation. I focus in particular on some warnings issued by the great teachers on these topics with respect to the motivations one ought to have in the Christian life. I then discuss some findings of contemporary psychology on habit formation which seem to be at odds with the warnings in Christian moral wisdom. I argue that while there is surface discord between (...) the contemporary psychology of habit formation and Christian moral wisdom, there is in fact a deep concord between them. (shrink)

Disorders of volition are often accompanied by, and may even be caused by, disruptions in the phenomenology of agency. Yet the phenomenology of agency is at present little explored. In this paper we attempt to describe the experience of normal agency, in order to uncover its representational content.

We discuss three varieties of affective dynamics. In each case, we suggest how these affective dynamics should be operationalized and measured in daily life using time-intensive methods, like ecological momentary assessment or ambulatory assessment, and recommend time-sensitive analyses that take into account not only the variability but also the temporal dependency of reports. Studies that explore how these affective dynamics are associated with psychological disorders and symptoms are reviewed, and we emphasize that these affective processes are within a nexus of (...) other components of emotion regulation. In this paper we defend the doxastic conception of delusions against the metacognitive account developed by Greg Currie and collaborators. According to the metacognitive model, delusions are imaginings that are misidentified by their subjects as beliefs: the Capgras patient, for instance, does not believe that his wife has been replaced by a robot, instead, he merely imagines that she has, and mistakes this imagining for a belief. We argue that the metacognitive account is untenable, and that the traditional conception of (...) delusions as beliefs should be retained. (shrink)

Quantum probability (QP) provides a new perspective for cognitive science. However, one must be clear about the outcome the QP model is predicting. We discuss this concern in reference to modeling the subjective probabilities given by people as opposed to modeling the choice proportions of people. These two models would appear to have different cognitive assumptions.

ABSTRACT:Drawing upon the collective action model of institutional change, I reconceptualize moral imagination as both a social process and a cognitive one. I argue that moral outcomes are not produced by individual actors alone; rather, they emerge from collective action processes that are influenced by political conditions and involve behaviors that include issue framing and resource mobilization. I also contend that individual moral imagination involves the integration of moral sensitivity with consideration of collective action dynamics. I illustrate my arguments with (...) a case study of the Chad-Cameroon oil project. The paper suggests new directions in teaching and research on moral imagination. (shrink)

Contemporary epistemology has been moving away from classical foundationalism—the thesis that our empirical knowledge is grounded in perceptual beliefs we know with certainty. McGrew reexamines classical foundationalism and offers a compelling reconstruction and defense of empirical knowledge grounded in perceptual certainty. He articulates and defends a new version of foundationalism and demonstrates how it meets all the standard criticisms. The book offers substantial rebuttals of the arguments of Kuhn and Rorty and demonstrates the value of the classical analytic approach to (...) philosophy. In this paper I critically examine the implications of the uncodifiability thesis for principlism as a pluralistic and non-absolute generalist ethical theory. In this regard, I begin with a brief overview of W.D. Ross’s ethical theory and his focus on general but defeasible prima facie principles before turning to 2) the revival of principlism in contemporary bioethics through the influential work of Tom Beauchamp and James Childress; 3) the widespread adoption of specification as a response to the indeterminacy of abstract (...) general principles and the limitations of balancing and deductive approaches; 4) the challenges raised to fully specified principlism by the uncodifiability thesis and 5) finally offer a defense of the uncodifiability thesis against various critiques that have been raised. She recalled how they saw things as composed of opposites; not torn, but (...) completed and made whole by them.Theirs was not a Manichean brand of dualism that sees matter, flesh, the body, as inherently evil, because they constantly addressed each other as “soully-gal”—soul: spirit; gal: the body, flesh, the visible self. And it was clear from their tone that they gave one as much weight and importance as the other. They had never heard of the mind/body split. 1This closing claim is surely less than self-evident. Whether or not the companions had heard of the mind/body split depends on the meaning given to the verb “hear.” Certainly it was embedded in their language, however much the phrase “soully-gal” might recombine its parts.Did the split come from Descartes and early European debates, linguistically colonizing all those who came within its sphere—Africans, Barbadians, Brooklynites? Did it come from a Christian heritage, with its haggling over bodily mortality and immortality of soul? Who knows? First in a tetralogy, this novel recounts something like a journey away from a life of colonial and postcolonial violence and oppression towards a hoped-for rebirth in “fulfilment and understanding.” 2 Early on, the harsh protagonist, Donne, mocks the narrator, his mysterious “double” and “brother,” Dreamer, who has remarked their distance from “the folk.” “Is it a mystery of language and address?” asks Donne. Slightly bewildered, Dreamer struggles to suggest that the obstacle is less of language than of a deeper disjunction:“it’s an inapprehension of substance,” I blurted out, “an actual fear of the substance of life, fear of the substance of the folk, a cannibal blind fear in oneself. Put it how you like,” I cried, “it’s a fear of acknowledging the true substance of life.
